pGEM®-T Easy . Parental vector for TA cloning of PCR products. The insertion site is flanked by BstZI, EcoRI, and NotI sites. To see this sequence with restriction sites, features, and translations, please download SnapGene or the free SnapGene .

pGEM®-T Easy (linearized) Linearized vector for TA cloning of PCR products. The insertion site is flanked by BstZI, EcoRI, and NotI sites. To see this sequence with restriction sites, features, and translations, please download SnapGene or the free SnapGene Viewer.

pGEM®-T. Parental vector for TA cloning of PCR products. The insertion site is flanked by BstZI sites. This vector is also known as pGEM®?5Zf (+). To see this sequence with restriction sites, features, and translations, please download SnapGene or the free SnapGene Viewer.

To see this sequence with restriction sites, features, and translations, please download SnapGene or the free SnapGene Viewer. pGEM – T (linearized).dna Map and Sequence File: Download Open, T-Overhangs for Easy PCR Cloning: The pGEM ®-T and pGEM -T Easy Vectors are linearized vectors with a single 3´-terminal thymidine at both ends. The T-overhangs at the insertion site greatly improve the efficiency of ligation of PCR products by preventing recircularization of the vector and providing a compatible overhang for PCR products, Addgene: Vector Database – pGEM-T Easy Vector, pGEM -T and pGEM -T Easy Vector Systems – Promega, pGEM-T Easy Sequence and Map, The only difference between pGEM-T and pGEM-T Easy is in the multiple cloning site (MCS). The MCS of the pGEM-T Easy Vector contains sequences on either side of the insert that are recognized by the restriction enzymes Not I and EcoR I. This allows the insert DNA to be removed with a single restriction digest using either of these enzymes.

Vector for standard cloning, efficient in vitro RNA synthesis, and ssDNA production. Identical to pGEM®?3Zf (+) except for the orientation of the f1 origin. To see this sequence with restriction sites, features, and translations, please download SnapGene or the free SnapGene Viewer.

Plasmid pGEM-T Easy -Gbait-hs-Cre from Dr. Marnie Halpern’s lab contains the insert Gbait-hsp70-Cre-SV40pA and is published in Unpublished This plasmid is available through Addgene. Image: Illustrated plasmid map in PNG format, Step 1: Clone the amplified Amy1 insert from from SnapGene Exercise #1 into pGEM – T easy vector Go to SnapGene program, click on “ File ” then “ Open ” to display the Amy1 intron sequence you created in SnapGene Exercise #1. Next, go to the “ Actions ” tab and from the pull-down menu select “ TA or GC cloning ” and then “ TA cloning ”.

In this exercise, you will use the DNA file you cloned and saved from Exercise #2 (i.e. “Cloned pGEM – T Easy +Intron Amy1”) and compare it to suspect 1 and culprit DNA sequences found on Canvas (CYSI SnapGene : CYSI SnapGene #3). Locate the file containing your cloned DNA from Exercise #2 and open it in SnapGene . Go to Canvas (CYSI SnapGene Exercises SnapGene #3) and click on both the …
